Composition:
Each tablet
contains:
Salbutamol ………………… 2 mg
Each 5 ml of syrup contains:
Salbutamol ………………… 2 mg
Properties:
Salbutamol is a B2
adrenergic drug that stimulates adenyl
cyclase the enzyme which catalyzes the
formation of cyclic AMP from ATP.The cyclic
AMP thus formed mediates the cellular
responses. Salbutamol exerts direct &
preferential action on beta 2
adrenoreceptors including those of the
bronchial tree & uterus.
Salbutamol is
readily absorbed from the G.I. tract. It is
subject to first - pass metabolism in the
liver, about a half is excreted in the urine
as an inactive sulphate conjugate, following
oral administration (the rest being
unchanged Salbutamol) The plasma half life
has been estimated to range from about 2 to
as much as 7 hours. In general the
intermediate values have followed oral
administration .
Indications:
For the relief
of bronchospasm in patients with reversible
obstructive airway disease e.g. bronchial
asthma,
bronchitis ………., etc.
Contra-indications:
The drug is
contraindicated in patients with a history
of hypersensitivity to Salbutamol.
Precautions:
Salbutamol should be used with caution in
patients with cardiovascular disorders, in
patients with hyperthyroidism or diabetus
mellitus & in patients who are unusually
responsive to sympathomimetic amines
Use during pregnancy & in nursing mothers:
► Salbutamol
should be used during pregnancy only if the
potential benefit justifies the potential
risk to the fetus.
► A decision
should be made whether to discontinue
nursing or to discontinue the drug, taking
into account the importance of the drug to
the mother.
Drug interactions:
The concomitant use of Salbutamol & other
oral sympathomimetic agents is not
recommended since such combined use may lead
to deleterious cardiovascular effects.
Salbutamol should be administered with
extreme caution to patients being treated
with M.A.O. inhibitors or tricyclic
antidepressants since the action of
Salbutamol on the vascular system may be
potentiated. Beta-receptor blocking agents &
Salbutamol inhibit the effect of each other.
Dosage and administration:
Adults:
One or two tablets 3 - 4 times
daily.
Elderly & sensitive patients:
One tablet 3 - 4 times daily.
Children:
Children Under 2 years:
100 micrograms / 1 kg / 4 times daily.
2 - 6 years :
I - 2 mg (half - one teaspoonful) 3 - 4 times daily
6 -12 years:
2 mg (one teaspoonful) 3 - 4 times daily.
Side effects:
Salbutamol may
cause fine tremors of skeletal muscles,
palpitations& muscle cramps Slight
tachycardia, tenseness, headache, peripheral
vasodilatation & hypokalaemia have been
reported after large doses
Overdosage:
Manifestations of over dosage include anginal pain, hypertension &
exaggeration of the effects listed in side effects. Treatment consists
of discontinuation of Salbutamol together with appropriate symptomatic
treatment.
