Composition:
Each capsule contains
Omeprazole.................. 20 / 40 mg
Properties:
Omepak
promotes healing of ulcers of the stomach,
duodenum, & esophagus.
It inhibits secretion of gastric acid by
irreversibly blocking the enzyme system of
hydrogen/ potassium adenosine triphosphate,
the so called the proton pump of the
gastric parietal cell.
It is valuable in those whose peptic ulcer
is not well controlled by H2 antagonists.
Omepak also enhances antibiotic activity
against H pylori by raising gastric pH.
Omepak is rapidly absorbed and is
extensively bound (about 95%) to plasma
protein. It is cleared by the circulation by
hepatic metabolism and most of the
metabolized are excreted in the urine.
Indications:
► Peptic ulcer
► Duodenal ulcer
►
Gastro-
Esophageal Reflux
►
Zollinger-Ellisson
syndrome
Use during pregnancy and lactation:
Omepak is used
during pregnancy only if the potential
benefit justifies the potential riskto the
fetus. Because of the potential for serious
adverse reaction in nursing infants from
Omepak a decision should be made whether to
discontinue nursing or to discontinue the
drug taking of course into account the
importance of the drug to the mother.
Dosage & route of
administration & duration:
► Peptic ulcer
and reflux oesophagitis
1 or 2 capsules daily as single dose for 4
to 8 weeks
► Zollinger-Ellisson
syndrome
3 to 4 capsules and maybe increased up to 9
capsules daily in divided doses in long term
treatment
Drug interaction:
Since Omepak
interacts with cytochrome P 450 in vitro,
inhibition of hepatic metabolism of certain
drugs may be important. Although the
clearance of diazepam is reduced by about
50%, Omepak has only minor effects on the
elimination of single oral doses of
phenytoin. It also increases the
anticoagulant activity of warfarin
Side effects:
Omepak is well
tolerated in treatment of patients with
Zollinger-Ellisson syndrome with doses of
180-360mg/ day for up to 4 years had not
caused any adverse or side effects.
Nausea, diarrhea, abdominal colic,
paraesthesia, dizziness and headache have
been stated to be generally mild, transient
and not requiring a reduction in dosage.
Storage:
Keep away from
heat, light, moisture, and at temperatures
not exceeding 30ºC
